Abstract. T cell epitope cloning targeting cellular immune response and serological analysis of recombinant cDNA expression libraries (SEREX) targeting humoral antibody response in cancer patients are the modern methods aimed on definition of cancer-associated antigens. We applied SEREX-analysis on few colon cancer cases in order to determine the tumor-derived antigens eliciting high-titer serum antibody response in tested patients. Totally, five different antigens designated as KY-CC-1-3, -21, -22 immunoreactive with autologous sera have been defined. Allogeneic immunoscreening of identified antigens with the set of sera samples from healthy donors and colorectal cancer patients revealed that KY-CC-3/COX1 antigen (previously reported as associated with autoimmune Hashimoto’s thyroiditis) could represent tumor-related antigen. We also suggest that implementation of plaque-spot serological assay should be a way to standardize allogeneic immunoscreening procedure — the most crucial step in SEREX analysis. Obstacles and possible ways to improve effectiveness of SEREX-procedure for colon cancer are discussed.
