EXPRESSION OF a- AND b-ISOFORMS OF ð70S6 KINASE IN HUMAN ENDOMETRIAL TUMORS

Abstract. Aim: to investigate the character of expression of p70S6Êa and b in human endometrial tumors and conventually normal tissues surrounding tumor. Methods: the expression of p70S6Êa and b has been studied in 16 samples of human uterine adenocarcinoma, 2 samples of adenosquamous cancer, and blood sera samples of mentioned patients and healthy donors by Western blot analysis, ELISA, immunohistochemistry, RT-PCR. Results: by Western blotting, hyperexpression of p70S6Êa and p70S6Êb was revealed in 56 and 67% cases of uterine adenocarcinoma compared to normal tissues. p70S6Êb mRNAs level was higher than that of p70S6Êa in tumor tissues and correlates with expression of kinase on protein level. Immunohistochemical analysis has shown the increased p70S6Êa expression in tumor stroma, endothelium of the vessels both of tumors and adjusting non-tumor tissues. Conclusion: p70S6Êa and b isoforms may play alternative roles in malignant transformation and progression of human endometrial tumors.
